Use and Management of the Sterile Room The sterile room shall be kept clean and orderly. Only essential laboratory tools—such as alcohol lamps, alcohol swabs, matches, forceps, inoculation needles, inoculation loops, and glass pens—may be stored inside; no items unrelated to the experiments are permitted. All inspection tools, workbenches, and stools must remain in their designated positions and may not be moved at will.
Before sterilization in the aseptic room, all items should be placed at the work station (except those awaiting inspection), after which the ultraviolet lamp is turned on for 30 minutes. Once the prescribed time has elapsed, the UV lamp is switched off and the room may be used. Prior to entering the aseptic room, personnel must change into disinfected lab coats, caps, and shoes in the buffer room. All procedures must be performed strictly in accordance with aseptic technique; minimize talking and avoid making noise during operations to maintain the sterility of the environment.
When aspirating bacterial suspension, use a rubber bulb; do not allow the pipette tip to come into direct contact with your mouth. If the pipette tip touches your hands or any other unsterilized surfaces, it must be replaced to prevent contamination. Before and after each use, the inoculation loop should be sterilized by flaming, and inoculation may proceed only after the culture has cooled.
Immerse pipettes, test tubes, and other glassware containing bacterial suspensions in a disinfection bucket containing 5% lysol solution for sterilization, and remove and rinse them after 24 hours. Before handling the culture medium, petri dishes must be re-sterilized in an autoclave. Do not flush these materials directly down the drain to avoid clogging. If bacterial suspension is spilled on a table or floor, immediately pour 5% sodium carbonate solution or 3% lysol over the contaminated area and allow it to act for at least 30 minutes before disposing of it. When work uniforms or caps become contaminated with bacterial suspension, they should be removed immediately, autoclaved, and then washed.
Preparation begins with disinfecting the aseptic room; the ultraviolet lamp should be turned on for 30–60 minutes. Prior to entering the aseptic room, all relevant testing equipment must undergo sterilization and disinfection. Before entering, personnel must thoroughly clean and disinfect both hands and don sterile work attire, including a cap and shoe covers. Upon entering the aseptic room, hands are disinfected again before proceeding with the examination procedures.
